Modern approaches to antiresorptive therapy for metastatic bone defeat (review of literature)

Summary. The purpose was to evaluate the effect of antiresorptive drugs on the incidence of skeletal related events on the example of comparing the treatment results of denosumab and zoledronic acid using the results of modern randomized trials. The articles were analyzed in PUBMED, EMBASE, Cochrane Library, Web of Science, databases from January 2012 to March 2018. Were included the results of four studies comparing the impact of the use of denosumab and zoledronic acid in bone metastases of various histological forms of tumors. The total number of patients was 11 087 of which 5551 patients received therapy with denosumab, and 5536 — with zoledronic acid. According to the results of the presented studies, the use of denosumab compared with zoledronic acid significantly reduces the risk of skeletal related events in patients with metastatic bone defect, regardless of the type of tumor, which in turn improves the quality of life and overall survival.
